Sporocadaceae is a species-rich and cosmopolitan fungal family including species of plant pathogens, endophytes or saprobes, and parasites of humans and animals. The taxonomy of Sporocadaceae has recently been revised using a polyphasic approach. However, much remains unknown about the diversity of species and their host associations. A collection of 488 strains, mostly from China and associated with 129 host plant species, was studied based on morphological comparisons and multi-locus (LSU, ITS, tef-1α , tub2 , and rpb2 ) phylogenies. Our results revealed that they belonged to 86 species, one new genus ( Cavernicola gen. nov .) and seven known genera, including Discosia , Monochaetia , Neopestalotiopsis , Pestalotiopsis, Seimatosporium , Seiridium and Sporocadus . Of these, 43 new species and three new combinations ( Dis . kaki , Mon . bulbophylli , and Neo . keteleeriae ) are proposed in this paper. In addition, Neo. vaccinii , Pes. kaki and Pes. nanjingensis are synonymised under Neo. hispanica , Pes. menhaiensis and Pes. sichuanensis , respectively. We also corrected seven problematic sequences of type materials of previously published species, namely Neo . iranensis ( tef-1α , ITS, tub2 ), Pes . jesteri ( tef-1α ), Pes . photinicola (ITS, tub2 ) and Pes . yunnanensis (ITS). Based on this study, Pestalotiopsis and Neopestalotiopsis are the most commonly detected genera within the Sporocadaceae family, associated with 84 and 70 plant species, respectively. Furthermore, considering the importance of Sporocadaceae and the fact that commonly used loci provide little valid information for species delimitation in this family, especially for Neopestalotiopsis and Pestalotiopsis , we initiated a phylogenomic project in this study. It will not only contribute to the knowledge of species boundaries but will also provide an important basis for evolutionary studies and research on secondary metabolites in Sporocadaceae . In this paper, a total of 26 pestalotioid isolates associated with different medicinal plants from southwest China and Thailand were studied. Based on morphological examinations and multigene analyses of three gene loci ( ITS , tef1-α , and tub2 ), these 26 isolates represent 17 species distributed in three genera, including seven new species and eight new records. The concatenated three loci tree was used to infer the occurrence of sexual recombination within each pestalotioid genus through the pairwise homoplasy index (PHI) test implemented in SplitsTree. Further, simplifying the description of pestalotioid species is discussed, and a checklist for pestalotioid species associated with medicinal plants worldwide is provided. IMPORTANCE Pestalotioid species are an important fungal group, occurring commonly as plant pathogens, endophytes, and saprophytes. The study of pestalotioid species associated with medicinal plants is significant for agriculture, industry, and pharmaceutical industry but remains poorly studied. In this study, we report 17 pestalotioid species related to medicinal plants based on morphology and molecular analyses. Our study significantly enriches the species richness of pestalotioids and provides a basis for follow-up studies.

Fagaceae is a family of flowering plants widely distributed in the Northern Hemisphere, including deciduous and evergreen trees and shrubs. Species of Pestalotiopsis are well-known agents of leaf spot diseases, but targeted sampling on Fagaceae is still missing. To determine the diversity of Pestalotiopsis species associated with Fagaceae leaf spot in China, investigations were conducted in the main areas of Fagaceae distribution from 2016 to 2021. Diseased leaf tissues were collected, and fungal isolates were obtained from leaf spots. In the present study, 43 isolates of Pestalotiopsis were studied based on combined morphology and phylogeny. As a result, 10 new species were identified, viz. , Pestalotiopsis anhuiensis , P. castanopsidis , P. changjiangensis , P. cyclobalanopsidis , P. foliicola , P. guangxiensis , P. guizhouensis , P. lithocarpi , P. shaanxiensis , and P. silvicola , and six new host records were recognized. IMPORTANCE Pestalotiopsis is a common fungal genus inhabiting plant tissues as endophytes, pathogens, and saprophytes. Fagaceae is a plant family including many important tree species, such as Castanea mollissima and Quercus spp. In this study, diseased leaves of Fagaceae in China were investigated, and 16 Pestalotiopsis species were identified based on morphology and phylogeny of combined loci of internal transcribed spacers (ITS), the translation elongation factor 1-α ( tef1 ), and the beta-tubulin ( tub2 ) genes. Among these, 10 new species were found, and six new host records were revealed. Our study significantly updates the taxonomy of Pestalotiopsis and enhances our understanding of leaf diseases of Fagaceae hosts.

Rosa ( Rosaceae ) is an important ornamental and medicinal plant genus worldwide, with several species being cultivated in China. Members of Sporocadaceae (pestalotioid fungi) are globally distributed and include endophytes, saprobes but also plant pathogens, infecting a broad range of host plants on which they can cause important plant diseases. Although several Sporocadaceae species were recorded to inhabit Rosa spp., the taxa occurring on Rosa remain largely unresolved. In this study, a total of 295 diseased samples were collected from branches, fruits, leaves and spines of eight Rosa species ( R . chinensis , R . helenae , R . laevigata , R . multiflora , R . omeiensis , R . rugosa , R . spinosissima and R . xanthina ) in Gansu, Henan, Hunan, Qinghai, Shaanxi Provinces and the Ningxia Autonomous Region of China. Subsequently 126 strains were obtained and identified based on comparisons of DNA sequence data. Based on these results 15 species residing in six genera of Sporocadaceae were delineated, including four known species ( Pestalotiopsis chamaeropis , Pes . rhodomyrtus , Sporocadus sorbi and Spo . trimorphus ) and 11 new species described here as Monochaetia rosarum , Neopestalotiopsis concentrica , N . subepidermalis , Pestalotiopsis tumida , Seimatosporium centrale , Seim . gracile , Seim . nonappendiculatum , Seim . parvum , Seiridium rosae , Sporocadus brevis , and Spo . spiniger . This study also represents the first report of Pes . chamaeropis , Pes . rhodomyrtus and Spo . sorbi on Rosa . The overall data revealed that Pestalotiopsis was the most prevalent genus, followed by Seimatosporium , while Pes . chamaeropis and Pes . rhodomyrtus were the two most prevalent species. Analysis of Sporocadaceae abundance on Rosa species and plant organs revealed that spines of R . chinensis had the highest species diversity. Citation : Peng C, Crous PW, Jiang N, et al. 2022. Tea-oil tree ( Camellia oleifera ) is an important edible oil woody plant with a planting area of over 3,800,000 hectares in southern China. Pestalotioid fungi are associated with a wide variety of plants worldwide along with endophytes, pathogens, and saprobes. In this study, symptomatic leaves of C. oleifera were collected from Guangdong, Guangxi, Hainan, Hunan, and Jiangsu Provinces and pestalotioid fungi are characterized based on combined sequence data analyses of internal transcribed spacer (ITS), beta tubulin ( tub2 ), and translation elongation factor 1-alpha ( tef-1α ) coupled with morphological characteristics. As a result, seven species were confirmed, of which five species are described as new viz. N. camelliae-oleiferae , P. camelliae-oleiferae , P. hunanensis , P. nanjingensis , P. nanningensis , while the other two are reported as known species, viz., N. cubana and N. iberica . Pathogenicity assays showed that all species except for P. nanjingensis developed brown lesions on healthy leaves and P. camelliae-oleiferae showed stronger virulence.
