Background The people in Ethiopia have developed their own specific knowledge to use, manage and conserve plant resources, giving traditional medicine its diverse nature. Documenting and investigating the traditional and cultural use of remedial plants is vital to extract bioactive chemicals and preserve plant species. This research was conducted with the aim of documenting ethnobotanical and associated knowledge on medicinal plants in Asagirt District, northeastern Ethiopia. Methodology The study was conducted from September 27, 2018, to April 9, 2019. A total of 367 informants (244 males and 123 females) were involved in the interviews. General informants (n = 349) were randomly selected, whereas key informants (n = 18) were selected purposively. Data were collected by using semistructured interviews, group discussions and guided field walks. We performed direct matrix ranking and preference ranking, and calculated the fidelity level and informant consensus factor (ICF). Descriptive statistics, including analysis of variance (ANOVA) and independent sample t-test were used to analyse the data. Results Overall, 103 medicinal plant species belonging to 96 genera under 45 plant families were recorded to be used by Asagirt people to alleviate different health problems. The species used to heal human diseases only were (64%, 66 species) followed by both livestock and human ailments (31%, 32 species) and livestock diseases only (5%, 5 species). Asteraceae and Fabaceae were best-represented (10.7%, 11 species each). The most frequently used plant parts were leaves (28%, 29 species), followed by seeds (16%, 17 species). The most important method of herbal remedy preparation was crushing (20.4%, 21 species). The common route of administration was oral (46.6%, 48 species), followed by dermal (22.3%, 23 species). Febrile illness, fever, headache, amoebiasis, typhoid and diarrhoea ailment categories had the highest ICF value (0.99). Ocimum lamiifolium Hochst. ex Benth. scored the maximum fidelity level value (98%). Conclusion Asagirt District is comparatively rich in medicinal plants and their associated knowledge. However, firewood collection, construction, the expansion of agricultural activities and timber production are the major challenges to medicinal plants. Hence, joint management with the people in Asagirt District in overall medicinal plant conservation would save medicinal plant resources.

Toad venom is regarded as the main source of bufadienolides; however, synthesis of these substances takes also place in a variety of other animal and plant organisms, including ethnomedicinal plants of the Kalanchoe genus. Chemically, bufadienolides are a group of polyhydroxy C-24 steroids and their glycosides, containing a six-membered lactone (α-pyrone) ring at the C-17β position. From the pharmacological point of view, bufadienolides might be a promising group of steroid hormones with cardioactive properties and anticancer activity. Most of the literature concerns bufadienolides of animal origin; however, the medicinal use of these compounds remains limited by their narrow therapeutic index and the risk of development of cardiotoxic effects. On the other hand, plants such as Kalanchoe are also a source of bufadienolides. Kalanchoe pinnata (life plant, air plant, cathedral bells), Kalanchoe daigremontiana (mother of thousands) and other Kalanchoe species are valuable herbs in traditional medicine of Asia and Africa. The present review focuses on the available data on chemical structures of 31 compounds, biological properties and prospects for therapeutic use of bufadienolides from Kalanchoe species. Furthermore, it presents some new investigational trends in research on curative uses of these substances.

Phenolic acid composition, antioxidant, and cytotoxic activities in leaves of four Kalanchoe (Crassulaceae) species were evaluated. Determination of phenolic acid contents were conducted by an optimized LC-ESI-MS/MS method. The results show that Kalanchoe daigremontiana Raym.-Hamet & H. Perrier (using ASE extraction) and Kalanchoe pinnata (Lam.) Pers. contain the highest amounts of phenolic acids, while Kalanchoe nyikae Engl. the lowest ones. Among phenolic acids ferulic, caffeic and protocatechuic acids were occurring in the highest quantities in the analysed species. The greatest amounts of ferulic and protocatechuic acids were found in K. daigremontiana and K. pinnata. Moreover, the antiradical and cytotoxic activities of Kalanchoe extracts were investigated. All tested extracts possessed antioxidant activity. The obtained IC 50 values (μg/mL) ranged from 49.9 μg/mL to 1410 μg/mL, indicating a large variation of the activity of the analysed extracts. Cytotoxicity assays revealed dose-dependent effects in the cells lines tested. Only K. pinnata extract showed a high cytotoxicity against the H-9 human T cell line. Other extracts ( K. daigremontiana , Kalanchoe milloti , K. nyikae ) showed more pronounced cytotoxicity towards J45.01 cells (human acute lymphoblastic leukaemia T cells). The present study demonstrated that Kalanchoe extracts have significant antioxidant and cytotoxic effects. This suggests that these species can be used as new sources of natural antioxidants and potential anticancer compounds.

Background Traditional medicines remained as the most affordable and easily accessible source of treatment in the primary health care system among diverse communities in Ethiopia. The Oromo community living in the prehistoric Harla and Dengego valleys has long history of ethnomedicinal know-how and practice against human and livestock ailments. However, this rich ethnomedicinal knowledge had been remained unexplored hitherto. This study focus on the comprehensive ethnomedicinal investigation in an attempt to safeguard the deteriorating ethnomedicinal knowledge that can be used as a stepping-stone for phytochemical and pharmacological analysis. Methods Fifty five (44 male and 11 female) systematically selected informants including ten traditional herbalists (key informants) were participated in the study. Semi-structured interviews, discussions and guided field walk constituted the data collection methods. Factor of informant consensus (Fic), frequency of citation (F%), and binomial test were employed in data analysis. Medicinal plant specimens were collected, identified and kept at Herbarium of Haramaya University (HHU). Results A total of 83 traditional medicinal plant species against human ailments in 70 genera and 40 Families were recorded. Twelve medicinal plants were marketable in open market places of the nearby towns. Formulations recorded added to 140 remedies for 81 human ailments. Concoction accounts 50.7% of the total preparations followed by fluids extraction (10.7%) and infusion (6.4%). Fifteen different plant parts were used for remedies preparation wherein leaves accounted 46.4%, stem 9.2%, fruits and roots each 7.8%. Most of the remedies (90.7%) were prepared from single plant species like, aphrodisiac fresh rhizome of Kleinia abyssinica (A. Rich.) A. Berger chewed and swallowed few hours before sexual performance for a man having problem of erectile dysfunction. The Fic value ranges between 1.0 (gastritis and heartburn/pyrosis) and 0.77 (swollen body part). Aloe harlana Reynolds was reported to be used for the highest number of ailments treating swollen body part locally called GOFLA, colon cleaner, snake bite, liver swelling, spleen swelling/splenomegaly, fungal infections and inflammation of skin. Conclusion Such documentation of comprehensive ethnomedicinal knowledge is very valuable and needs to be scaled-up so that it could be followed up with phytochemical and pharmacological analyses in order to give scientific ground to the ethnomedicinal knowledge.

Plant somatic cells have the remarkable ability to regenerate an entire organism. Many species in the genus Kalanchoë, known as "mother of thousands," develop plantlets on the leaf margins. Using key regulators of organogenesis (STM) and embryogenesis (LEC1 and FUS3) processes, we analyzed asexual reproduction in Kalanchoë leaves. Suppression of STM abolished the ability to make plantlets. Here, we report that constitutive plantlet-forming species, like Kalanchoë daigremontiana, form plantlets by coopting both organogenesis and embryogenesis programs into leaves. These species have a defective LEC1 gene and produce nonviable seed, whereas species that produce plantlets only upon stress induction have an intact LEC1 gene and produce viable seed. The latter species are basal in the genus, suggesting that induced-plantlet formation and seed viability are ancestral traits. We provide evidence that asexual reproduction likely initiated as a process of organogenesis and then recruited an embryogenesis program into the leaves in response to loss of sexual reproduction within this genus.
