Background DNA barcoding have been considered as a tool to facilitate species identification based on its simplicity and high-level accuracy in compression to the complexity and subjective biases linked to morphological identification of taxa. MaturaseK gene (MatK gene) of the chloroplast is very vital in the plant system which is involved in the group II intron splicing. The main objective of this study is to determine the relative utility of the "MatK" chloroplast gene for barcoding in 15 legume as a tool to facilitate species identification based on their simplicity and high-level accuracy linked to morphological identification of taxa. Methods and results MatK gene sequences were submitted to GenBank and the accession numbers were obtained with sequence length ranging from 730 to 1545 nucleotides. These DNA sequences were aligned with database sequence using PROMALS server, Clustal Omega server and Bioedit program. Maximum likelihood and neighbor-joining algorithms were employed for constructing phylogeny. Overall, these results indicated that the phylogenetic tree analysis and the evolutionary distances of an individual dataset of each species were agreed with a phylogenetic tree of all each other consisting of two clades, the first clade comprising (Enterolobium contortisiliquum, Albizia lebbek), Acacia saligna, Leucaena leucocephala, Dichrostachys Cinerea, (Delonix regia, Parkinsonia aculeata), (Senna surattensis, Cassia fistula, Cassia javanica) and Schotia brachypetala were more closely to each other, respectively. The remaining four species of Erythrina humeana, (Sophora secundiflora, Dalbergia Sissoo, Tipuana Tipu) constituted the second clade. Conclusion Moreover, their sequences could be successfully utilized in single nucleotide polymorphism or as part of the sequence as DNA fragment analysis utilizing polymerase chain reaction in plant systematic. Therefore, MatK gene is considered promising a candidate for DNA barcoding in the plant family Fabaceae and provides a clear relationship between the families.

The taxonomy and evolutionary history of Sophora L., a genus with high economic and medicinal value, remain uncertain due to the absence of genetic resource (especially in China) and low polymorphism of molecular markers. Our aim was to elucidate the molecular evolution and phylogenetic relationships in chloroplast genomes of Sophora species in the early-diverging legume subfamily Papilionoideae (Fabaceae). We reported nine Sophora chloroplast genome from China using Illumina sequencing. We performed a series of analyses with previously published genomes of Sophora species to investigate their genomic characteristics, identified simple sequence repeats, large repeat sequences, tandem repeats, and highly polymorphic loci. The genomes were 152,953-158,087 bp in length, and contained 111-113 unique genes, including 76-78 protein coding, 31 tRNA, and 4 rRNA. The expansion of inverted repeat boundary of Sophora resulted in rps12 entering into the LSC region and loss of trnT - CGU gene in some species. Also, we found an approximately 23 kb inversion between trnC-GCA and trnF-GAA within the genus. In addition, we identified seven highly polymorphic loci (pi (π) > 0.035) suitable for inferring the phylogeny of Sophora species. Among these, three regions also co-occurred with large repeat sequences and support use of repeats as a proxy for the identification of polymorphic loci. Based on whole chloroplast genome and protein-coding sequences data-set, a well-supported phylogenetic tree of Sophora and related taxa showed that this genus is monophyletic, but sect. Disamaea and sect. Sophora , are incongruent with traditional taxonomic classifications based on fruit morphology. Our finding provides significant genetic resources to support further investigation into the phylogenetic relationship and evolution of the genus Sophora .

Gymnocladus chinensis Baill (Fam.: Leguminosae; Trib.: Caesalpinieae) are widely distributed in China. In this study, we assembled the complete chloroplast (cp) genome of G. chinensis . The total cp genome size was 165,315 bp in length, containing a large single-copy region of 92,356 bp, a small single-copy region of 20,449bp, and a pair of inverted repeat regions of 26,255 bp. The all GC content of G. chinensis cp was 34.95%. It encodes a total of 105 unique genes, including 75 protein-coding genes, 26 tRNA genes, and four rRNA genes. Seventeen genes contain a single intron, and two genes ( ycf3 and clpP ) have two introns. Phylogenetic analysis results strongly supported that G. chinensis was closely related to Angylocalyx braunii.

The inadequacy of information impedes society's competence to find out the cause or degree of a problem or even to avoid further losses in an ecosystem. It becomes even harder to identify all the biological resources at risk because there is no exhaustive inventory of either fauna or flora of a particular region. Coastal forests of Kenya are located in the southeast part of Kenya and are distributed mainly in four counties: Kwale, Kilifi, Lamu, and Tana River County. They are a stretch of fragmented forests ca. 30-120 km away from the Indian Ocean, and they have existed for millions of years. Diversity of both fauna and flora is very high in these relicts and the coastal forests of Eastern Africa, extending along the coast from Somalia through Kenya and Tanzania to Mozambique, are ranked among the priority biodiversity hotspot in the world. In spite of the high plant species richness and their importance towards supporting the livelihoods of the communities that live around them, floristic studies in these forests have remained poorly investigated. Hence, based on numerous field investigations, plant lists from published monograph/literature, and data from BRAHMS (Botanical Records and Herbarium Management System) database at East African herbarium (EA), we present a detailed checklist of vascular plants recorded in this region. Our results show that Kenyan coastal forests play an essential role in the flora of Kenya and the plant diversity of the coastal forests of East Africa. The checklist represents 176 families, 981 genera, 2489 species, 100 infraspecific taxa, 90 endemic plants species, 72 exotic species, and 120 species that are included in the current IUCN Red List of Threatened Species as species of major concern. We also discovered three new species to the world from these relicts. Thus, Kenyan coastal forests present a remarkable and significant center of plant diversity.

Background and aims The study of floral morphology and ontogeny and the re-investigation of existing data help to uncover potential synapomorphic characters and foster our understanding of phylogenetic relationships that rely primarily on molecular analyses. Goniorrhachis marginata is a monotypic caesalpinioid legume (Leguminosae) that shows some interesting floral features, such as a long hypanthium and regular Rosaceae-like flowers. We studied the ontogeny and morphology of the flowers in detail and present our results in a broad phylogenetic context. Methods Flower buds were collected in the field, fixed in 70 % ethanol and investigated using scanning electron microscopy. Older buds in spirit were carefully opened to investigate the direction of style bending. Characters of the style from 131 taxa from the main legume lineages were analysed and mapped on a Bayesian molecular phylogeny. Key results The tetramerous calyx is the result of complete loss of one sepal. The formation of the radially symmetrical corolla starts in a typical caesalpinioid pattern with the adaxial petal innermost (ascending aestivation). The young style bends in the abaxial direction, which is a character found exclusively in all studied detarioid legumes and therefore a newly described synapomorphy for the clade. Conclusions We show that investigation of unstudied taxa and reinvestigation of published data can uncover new, previously overlooked and important characters. Curvature of the style can be detected in young buds with a hand lens and therefore is an important character for field botanists. Our study reveals the importance of including poorly studied and/or phylogenetically enigmatic taxa in molecular phylogenies and in detailed morphological and ontogenetic analyses.
